Golgi α‐mannosidase II (GMII) is a key enzyme in the N‐glycosylation pathway and is a potential target for cancer chemotherapy. The natural product swainsonine is a potent inhibitor of GMII. In this paper we characterize the binding of 5α‐substituted swainsonine analogues to the soluble catalytic domain of Drosophila GMII by X‐ray crystallography. These inhibitors enjoy an advantage over previously reported GMII inhibitors in that they did not significantly decrease the inhibitory potential of the swainsonine head‐group. The phenyl groups of these analogues occupy a portion of the binding site not previously seen to be populated with either substrate analogues or other inhibitors and they form novel hydrophobic interactions. They displace a well‐organized water cluster, but the presence of a C(10) carbonyl allows the reestablishment of important hydrogen bonds. Already approximately tenfold more active against the Golgi enzyme than the lysosomal enzyme, these inhibitors offer the potential of being extended into the N‐acetylglucosamine binding site of GMII for the creation of even more potent and selective GMII inhibitors. 相似文献

Electrochemical double‐layer capacitors (EDLCs) are devices that store enormous amounts of charge electrostatically when a potential is applied between electrodes of very high surface area (typically made of porous carbon) and an electrolyte. Wider commercialization of this technology has been held back by the lack of ultralow‐cost electrode materials. We demonstrate that used coffee grounds can be processed to form low‐cost electrodes. The surface and electrochemical characteristics of microporous activated carbons from used coffee grounds (CGCs) were measured. First, optimal times and temperatures for carbonization and activation were identified on the basis of Brunauer–Emmett–Teller (BET) surface area, pore volume, and pore size distribution. Second, CGCs were used as polarized electrodes in EDLCs, whose capacitances were evaluated using cyclic voltammetry. The results show that carbonization for 1 h at 600 °C with a heating rate of 300 °C/h, followed by CO2 activation for 2 h at 1000 °C, affords the highest BET surface area (1867 m2/g) compared to other works. The produced CGCs have many micropores of less than 2 nm across, which contribute to the formation of an electric double layer. Capacitors made using these CGCs show the highest capacitance (103 F/g) in 0.8 M (C2H5)4NBF4/PC as an organic electrolyte, which is much higher than the ∼80 F/g typically used in organic‐electrolyte‐based commercial EDLCs, suggesting that coffee grounds are a useful electrode material. © 2014 Institute of Electrical Engineers of Japan. This article is concerned with the dynamic treatment of thermally induced stress waves in an infinite elastic plate subjected to impulsive electromagnetic radiation. The plate is assumed to be a functionally graded material (FGM), meaning that the material is composed of multiconstituents in ceramics and metals, the volume fractions of which distribute continuously inside the material. The mathematical problem is one of wave propagation in a typical nonhomogeneous material The radiation absorption is assumed to occur at a constant rate for the duration of the pulse and to diminish exponentially with distance from the surface of the plate, assuming negligible heat conduction. In treating problems, the nature of the stress-wave buildup in the plate is studied for the case of a temperature-dependent solid, that is, when material properties vary with temperature. The numerical procedure employs the characteristic method based on the integration of the governing equations along the characteristics. Numerical calculations are carried out for ceramic-metal FGM plates showing the influences of the temperature-dependent material properties and the volume fractions of the phases composing the FGM on the magnitude of the dynamic thermal stresses. 相似文献

In photoautotrophic cultures of pak-bung hairy roots, strong light irradiation (22 W/m2) increased the content of reactive oxygen species (ROS) in the cells, resulting in chlorophyll (Chl) degradation and DNA injury. The Chl degradation rate, RD, can be used as a parameter to measure the cellular damage caused by photo-induced stress. Pharmaceutical manufacturing plants can be operated continuously for several months. It is therefore important to use cells with long-term stability for the production of active ingredients. We investigated the reliability and long-term stability of an antibody-producing cell line. A recombinant Chinese hamster ovary (CHO) cell line was cultivated in spinner flasks and reactors, including a practical production-scale reactor (1600 L), for 109 days to produce monoclonal antibodies against the HM1.24 antigen. During cultivation, the cells remained stable and there was an increase in the rate of cell proliferation, yielding viable cells at high density. A decrease in cell-specific productivity was associated with this increase in the rate of cell proliferation. The cells were genetically stable and other measures of cellular function remained consistent throughout the cultivation period. 相似文献

In this study, we report the characterization of carbonaceous films deposited on metal substrates by liquid-phase electrodeposition in methanol. The characterization of carbonaceous films by electrodeposition was examined by means of Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), electron energy loss spectroscopy (EELS), secondary ion mass spectrometry (SIMS), atom probe (AP) and high resolution-elastic recoil detection analysis (HR-ERDA). From these results, it was found that the films deposited on the metal substrates were composed of the sp2 and sp3 carbon contents, of which the ratio was about 7:3. Furthermore, the films by electrodeposition contained much hydrogen. The hydrogen contents in the surface were about 60 at.% and those in the subsurface were a few 10 at.%. 相似文献

The Film Dryout Analysis Code in Subchannels, FIDAS, has been developed with the main objective of predicting dryout and post-dryout heat transfer in a channel and in rod bundles. In FIDAS, two-phase flow consisting of continuous liquid film, continuous vapor and entrained droplets is modeled by a three-fluid, three-field representation of 12 field equations, i.e. three continuity, three energy and six momentum equations. FIDAS can predict dryout without any empirical CHF correlations by introducing annular flow modeling and the ‘film dryout criterion’. Experiments on film flow characteristics, subchannel flow and enthalpy distributions, dryout and post-dryout heat transfer in tubes and rod bundles were analyzed to demonstrate the performance of FIDAS.
